烧伤痂下水肿液对内皮细胞单层通透性的影响 被引量:4

Effect of Subcrustal Tissue Fluid on Endothelial Cell Monolayer Permeability Under Hydrostatic Perfusion

摘要 目的 了解痂下水肿液 (STF)对内皮细胞通透性的影响。方法 应用体外灌注内皮细胞 (EC)单层通透性方法 ,测定内皮单层流出液速度 (Jv)和滤过数 (kf)及对蛋白质渗透压反射系数 (б)的变化 ,观察 10例烧伤病人早期痂下水肿液对人脐静脉内皮细胞 (HUVEC)单层通透性的影响 ,以及对HUVEC细胞活力及形态的改变。结果 烧伤病人早期STF可导致体外培养的EC细胞活力降低、细胞收缩、细胞间隙增大 ,内皮单层kf、Jv升高 ,б降低。结论 STF可损伤内皮细胞并可导致细胞通透性增高。 Objective To investigate the effect of subcrustal tissue fluid (STF) on the endothelial cell (EC). Methods The vascular permeability of perfused endothelial cell monolayer cultured on micropore filter membrane was studied to determinate the filtration coefficient (kf), transendothelial cell monolayer volume flux (Jv), and osmotic reflection coeficient (б) after EC was incubated with STF for 24 hours. Furthermore, the change of cell activity was determined and morphology was observed. Results Incubation of EC with 20% STF for 24 hours decreased the activity of human umbilical vein endothelial cell (HUVEC), shrinked HUVEC, enlarged intercellular space and increased permeability of endothelial cell monolayer. Conclusions STF can damage endothelial cells and induce increase of EC monolayer permeability.
