鼻咽癌患者外周血全基因组表达谱芯片结果初步分析 被引量:1

Primary analysis of nasopharyngeal carcinoma patient's peripheral blood whole-genome microarray

摘要 目的:从外周血水平比较健康对照者及鼻咽癌患者的外周血基因表达谱差异,寻找新的mRNA分子标志。方法:选取20例鼻咽癌患者和20名健康对照者进入本研究,两组性别、年龄基本匹配。采用QIAGENRNeasy○RMini Kit分离纯化外周血mRNA,行Agilent人类全基因寡核苷酸芯片检测。利用Gene Spring 10.0软件采用t-test une-qual unpaired算法,计算P<0.05时的差异基因并按差异倍数排序。利用Gene spring软件及DAVID Bioinformatics Resources 6.7对前期筛选的差异基因进行聚类分析,GO分析、Pathway分析,分析差异基因功能分类及相关通路。结果:差异倍数≥1.5倍且P<0.05的包含1 257个探针(上调687个,下调570个);差异倍数≥2.0倍的包含171个探针(上调132个,下调39个)。差异基因主要与细胞周期、免疫反应及对病毒的反应等生物学过程有关;主要参与肿瘤信号通路、细胞周期通路和p53信号通路等。结论:用全基因组表达谱芯片可以筛选出鼻咽癌患者与健康对照者外周血中的差异表达基因,进一步筛选这些基因可能寻找到鼻咽癌诊断相关的mRNA分子标志。 OBJECTIVE: To compare wholegenome expression differences between healthy control and nasopharynged carcinoma patient, and screen mRNA molecular biomarkers in periph eral blood of nasopharyngeal carcinoma patients. METHODS: Twenty cases of nasopharyngeal carcinoma (NPC) patients and 20 cases of healthy control which were matched in sex and age, were entered the study, mRNA was extracted from whole periph eral blood sample by QIAGEN RNeasy~ Mini Kit and then using Agilent gene oligonucleotide microarray to detect differentially expressed genes between the two groups. Using ttest unequal unpaired algorithm of Gene Spring 10.0 software to calculate dif ferentially expressed genes when P ~ 0. 05 and sort by fold change. Gene Spring and DAVID Bioinformatics Resources 6. 7 were used for clustering, GO analysis, Pathway analysis; Classi fication and function of different genes were illustrated. RE SULTS: For fold change larger than 1.5, there 1 257 probes (up reg ulation 687, down regulation 570); for fold change larger than 2.0, there are 171 probes (up regulation 132, down regulation 39). They were mostly related to cell cycle, immune response and reaction to the virus. They were mainly involved in cancer pathway, cell cycle path way, p53 signaling pathway. CONCLUSIONS: Wholegenome mi croarray can detect differentially expressed genes in peripheral blood between NPC patients and healthy control, mRNA molecular biomar kers should be found after deeper analysis.
