摘要
目的:分离克隆测序胃癌高表达基因gcys18 的部分c D N A. 方法:采用m R N A 差异展示技术从胃癌 G C7901 与胃粘膜 G E S1 细胞株中分离出一差异表达基因片段,对其命名,并进行克隆、测序及 Gen Bank 检索;以此片段作探针与细胞株总 R N A 进行打点杂交. 结果:从差示 P C R 电泳凝胶中分离出大小为416 bp 的差异表达片段,命名为gcys18; R N A 打点杂交证实gcys18 在 G C7901 中呈高表达,在 G E S1 中呈低表达,表明gcys18 是真正差异表达片段;对gcys18 进行了序列分析及 Gen Bank 检索,示其为新序列后递交给 Gen Bank,它的 Gen Bank 接受号为 A F071057. 结论:分离并克隆了胃癌高表达基因gcys18 的部分c D N A,此基因可能与胃癌发生发展有关.
AIM: To isolate, clone and sequence part cDNA of gcys 18 overexpressed in gastric carcinoma. METHODS: gcys 18, isolated from differential display gel between GC7901 and GES 1 by mRNA DD PCR, was cloned into T vector. As a probe, gcys 18 was hybridized to total RNAs of GC7901 and GES 1. It was then sequenced and searched against GeneBank. RESULTS: gcys 18 was isolated, cloned and found to be overexpressed in GC7901 and downexpressed in GES 1 by RNA dot blot. By sequencing, gcys 18 was confirmed to be 416 bp, partly similar to that of HEK5, and its accepted number in GeneBank is AF071057. CONCLUSION: Part cDNA of gcys 18 is isolated and cloned and it may be an important overexpressed gene in gastric carcinoma.
