摘要
目的:观察地白忍煎剂( D B R D)对角朊细胞和成纤维细胞增殖的影响以及对成纤维细胞收缩功能的潜在作用.方法:将人角朊细胞和真皮成纤维细胞分别培养于含不同稀释度 D B R D 的培养基中,以噻唑蓝染色法测定细胞增殖率.采用体外胶原立体培养模型观察 D B R D 作用后成纤维细胞收缩功能的变化. 结果:1∶107 稀释的 D B R D对角朊细胞和成纤维细胞均具有促增殖作用,而1∶10 稀释的 D B R D 仅促进角朊细胞的增殖. D B R D 剂量依赖性地抑制含成纤维细胞的胶原凝胶块的收缩率,且该抑制作用可被仅含100 m L/ L胎牛血清的培养基逆转. 结论: D B R D可促进角朊细胞和成纤维细胞的增殖。
AIM: To investigate the effects of dibairen decoctum (DBRD) on the proliferation of keratinocytes and fibroblasts as well as the potential effects of DBRD on fibroblast contraction. METHODS: Human keratinocytes and dermal fibroblasts were cultured in media containing serial dilutions of DBRD,which was followed by cell proliferation determination assessed with MTT assay at different time intervals. The in vitro collagen lattice contraction model was utilized to determine the contractility of fibroblasts cultured in medium containing DBRD. RESULTS: The 1∶10 7 dilution of DBRD enhanced the growth of both keratinocytes and fibroblasts whereas the 1∶10 dilution only increased the growth of keratinocytes. Collagen lattice contraction was inhibited dose dependently by DBRD and such an inhibition could be reversed by switching DBRD containing medium to normal medium containing 100 mL/L fetal calf serum. CONCLUSION: DBRD enhances the growth of keratinocytes as well as fibroblasts, and reversibly inhibits the fibroblast contraction in collagen lattice.
