摘要
目的以人成纤维细胞为载体对三种皂苷Rg1、Rb1、Re进行研究。方法 MTT法测定三种药物在不同浓度下对人成纤维细胞增殖的影响,消化法检测羟脯氨酸含量。ELISA法测定细胞分泌Ⅰ型前胶原、基质金属蛋白酶-1(matrix metalloproteinase-1,MMP-1)、基质金属蛋白酶抑制酶-1(tissue inhibitor of metalloproteinase-1,TIMP-1)的蛋白表达情况,RT-PCR法观察上述三种蛋白的mRNA表达水平。结果与空白组比较,三种单体均能显著提高细胞增殖水平,羟辅氨酸含量、Ⅰ型前胶原的蛋白质及mRNA表达量,显著降低MMP-1蛋白质及mRNA表达量。结论人参皂苷Rg1、Re、Rb1通过影响成纤维细胞的基因及蛋白质的表达,均显著促进胶原合成、抑制胶原降解,从而提高胶原蛋白总量,改善胶原代谢。
Objective Ginsenosid plays an important role in anti-aging of skin cells. The effects of ginsenoside monomers on collagen metabolism and its mechanism, however, have not been reported. To investigat the effect of ginsenoides Rgl , Rbl and Re on human skin collagen metabolism and its mechanism. Methods The proliferation of fibroblasts treated by different concentrations of three ginsenoides was measured with MTT method. The content of hydroxyproline was detected by digestive assay. The expressions of procollagen I , MMP-1 and TIMP-1 proteins and mRNA were measured by ELISA and RT-PCR respectively. Results Compared with the control group, the treatment of three ginsenosides significantly enhanced proliferation of fibroblasts, increased the amount of hydroxyproline, and expression of both protein and mRNA of procollagen I and TIMP-1, but significantly decreased expression of MMP-1 protein and mRNA. Conclusion Ginsenosides, Rgl, Re and Rbl can promote synthesis and suppress degradation of collagen in skin fibroblasts, which may be associated with regulating the mRNA and protein levels of procollagen I , MMP-1 and TIMP-1.
