奥曲肽对高脂诱导不同程度肥胖小鼠自由基和血糖的影响

摘要 目的:研究奥曲肽对高脂诱导不同程度肥胖小鼠自由基(ROS)和血糖(BG)的影响。方法:雄性C57BL/6小鼠80只,分为正常(control)、高脂(HFD)两组,饲喂5w后将HFD组分为肥胖(Obesity,OB)、高脂中体重(Middle Weight,MW)、肥胖抵抗(Obesity Resistance,OR)三组,每组各取8只分别腹腔注射奥曲肽(OCT,20μg/kg)和0.9%生理盐水0.1mL,注射10min后小鼠断尾取血测ROS和BG。小鼠称重后处死,取腹部脂肪称重并计算腹脂率。结果:饲喂5w后,与正常组相比,OB组小鼠体重显著增加,OR组显著降低(p<0.05),OB、MW、OR组小鼠腹脂重、腹脂率均显著增加(p<0.05);OR组小鼠血ROS与正常组相比显著升高(p<0.05),注射OCT之后,各组小鼠血ROS与注射NS相比均有所降低(p>0.05);OR、MW、OB组小鼠BG与正常组相比显著升高(p<0.05),注射OCT之后,OB、MW、OR组小鼠BG与注射NS相比均显著下降(p<0.05)。结论:饲喂5w的高脂饲料后小鼠出现了肥胖和肥胖抵抗,血ROS升高,其中肥胖抵抗组ROS显著升高,BG显著升高,OCT通过降低血ROS水平而显著降低高脂饲养组小鼠BG。 Objective:To study the effects of octreotide on free radicals and blood glucose of various degrees of obesity induced by high-fat diet in mice Method= 80 of Male C57BL/6 mice were assigned to two groups, the control group( n = 16)were administrated with an ordinary diet ;the HFD group( n = 64)were fed with high fat diet, after .5 weeks feeding,the HFD group were divided into three groups:the OB group (Obesity, n = 16, the top :25% body weight) ,the MW group ( Middle Weight, n = 32, the middle 50% of body weight), the OR group ( Obesity Resistance,n = 16,the last 25% body weight),8 mice in each group were injected by intraperitoneal injection of octreotide(OCT,20μg/kg) and 0.9% saline 0.1 mL, 10min after injection ,free radical(ROS) and blood glucose(BG) were determined.After weighting, all the mice were sacrificed by being broken cervical vertebrae, the weights of abdominal fat in were recorded and the abdominal fat percentages were calculated.Results:After 5 weeks feeding, compared with the normal group,The body weight of mice in OB group significantly increased,and the OR group significantly decreased(p 〈 0.05), the abdominal fat weight and fat rate in OB, MW and OR group significantly increased compared with the normal group ( p 〈 0.05) ; the blood ROS level of mice in OR group significantly increased compared with the normal group ( p 〈 0.05), after injection of OCT, the blood ROS level decreased compared with injection with NS ( p 〉 0.05) ; the BG of mice in OR, MW and OB group significantly increased compared with the normal group(p 〈0.05) ,the blood ROS level decreased compared with injection with NS( p 〉 0.05).After injection of OCT,The BG of mice in OB,MW and OR group significantly decreased Compared with the injection of NS( p 〈 0.05).Conclusion :5 weeks high-fat diet leads obesity and obesity resistance to mice. Increase ROS level of whole blood, with significantly increase in obesity- resistant group, significantly increase inBG,injection of OCT significantly decrease BG.Level by reducing the level of blood ROS in mice fed with high-fat diet.
