摘要
提出了一种测定黑松花粉破壁率的新方法。运用超声波破壁技术对黑松花粉破壁处理,根据超声波工作次数的差异获取不同的花粉破壁率,并借用分光光度法测出相同质量的花粉在不同超声波工作次数下的脯氨酸吸光度,同时显微镜下镜检其相应花粉的破壁率。结果表明:花粉中脯氨酸吸光度与其破壁率正相关。该曲线线性关系良好,准确度高,简单易操作,本方法为检测花粉破壁率提供了一项快速有效的手段。
A new method for assaying the rate of wall-broken pollens was put forward.Ultrasonic broken instrument was used to break the wall of Pinus thunbergii pollens and Spectrophotometer was used to measure the absorbance of Proline .After measuring the rate of wall-broken pollens, the standard curve of the absorbance of Proline and the rate of wall-broken pollens was received.The result indicated that it was a summary method with a good linear relationship.It makes the way of measuring the rate of wall-broken pollens easier,faster and preciser.
