海参多糖硫酸根含量的离子色谱法测定及其差异分析 被引量:10

Determination of sulfate content of sea cucumber polysaccharides by ion chromatography and analysis of the differences

摘要 经过三氟乙酸水解后,利用Dionex ICS-2000离子色谱仪对不同海参粗多糖、海参硫酸软骨素(SC-CHS)和海参岩藻聚糖硫酸酯(SC-FUC)中的硫酸根(SO42-)进行了分离和测定。平均加标回收率为100.90%,相对标准偏差为2.12%,检测限达到3.9×10-2μg/mL。较之传统的多糖硫酸根的测定方法,本方法重复性好,测定更为简便、准确,可应用于其它硫酸多糖硫酸根的定量测定。测定结果显示,所有种类海参SC-CHS的硫酸根含量高于SC-FUC,不同种类海参的多糖硫酸根含量存在特征性差异。 Sulfate in sea cucumber polysaccharide ( crude sample), sea cucumber chondroitin sulfate ( SC- CHS ) and sea cucumber fucan(SC-FUC)were hydrolyzed by TFA,separated and determinated by ICS-2000 system. The average recovery was 100.90%, RSD =2.12% ,the detection limitation reached 3.9 ×10^-2μg/mL.Compared with the traditional determination method of sulfate, this method was reproducible, much simple and more accurate.The method applied to quantitative determination of sulfate in other sulfated polysaccharides.The results showed that sulfate in all kinds of SC-CHS had a higher content than in SC-FUC,characteristical differences of sulfate content existed among different kinds of sea sucumber polysaccharide.
