摘要 目的:寻找卒中型自发性高血压大鼠卒中相关基因的遗传标记。方法和结果:用60 个随机引物对自发性高血压大鼠(SHR)和卒中型自发性高血压大鼠(SHRsp)两个品系基因组DNA进行随机扩增多态性分析(Random Am plified Polym orphicDNAs,RAPD),找到一个仅存在于SHRsp 而不存在于SHR的特异性DNA片段,此片段长630bp,命名为SHRsp-630。应用SHRsp-630 为探针,经Southern blot杂交证实,该序列在SHRsp 为强阳性而在SHR为弱阳性。经测序和GenBank 查询,证实SHRsp-630为新序列,GenBank 序列编号为G38021。结论:SHRsp-630 可能是与卒中相关基因紧密连锁的RAPD标记。 Aim:To search for the genetic markers linked to strok related genes in SHRsp. Methods:Using 60 random primers, genomic DNAs from SHR and SHRsp were analysed by RAPD (Random Amplified Polymorphic DNAs) technology. Results:One DNA fragment existing only in SHRsp, but not in SHR, was identified. This DNA fragment was 630 bp, and named SHRsp 630. Strong positive hybridization signal was found in SHRsp, and weak positive hybridization signal in SHR, by using SHRsp 630 as an hybridization probe on Southern blot. DNA sequence analysis and GenBank inquery confirmed that SHRsp 630 is a novel DNA fragment. The GenBank accession number is G38021. Conclusion:SHRsp 630 may be a RAPD marker linked to stroke related genes.
