摘要
目的探讨CD133+结直肠肿瘤干细胞表面ABC转运蛋白的表达。方法运用流式细胞仪(FACS)分析结直肠肿瘤细胞中CD133的表达,通过实时荧光定量PCR分析9种常见ABC转运蛋白在结直肠肿瘤细胞及正常组织细胞中的表达情况,并筛选出在肿瘤组织中高表达的ABC转运蛋白做免疫荧光分析,观察CD133+细胞中ABC转运蛋白的表达。结果有(2.59±1.26)%的结直肠肿瘤细胞表达CD133表面标记,在9种ABC转运蛋白中,MRP1在结直肠肿瘤细胞中表达显著高于正常组织细胞,共聚焦显微分析后证明CD133+细胞为主要MRP1表达细胞。结论 MRP1为结直肠肿瘤中主要表达的ABC转运蛋白,并且这一转运蛋白集中表达在CD133+肿瘤细胞中。
[Objective] To investigate the expression and distribution of ABC transporters in CD133 positive human eoloreetal cancer stem cells. [Methods] The expression of CD133 in coloreetal cancer ceils was analyzed by flow cytometry (FACS). Expression of ABC transporters was analyzed for tumor and normal control tissues by real-time fluorescence quantitative PCR. Those ABC transporters that were highly expressed in tumor tissues were selected and further analyzed for expression in CD133^+ and CD133- tumor cells. [Results] In fresh colorectal cancer Tissues, 2.59% CD133 positive ceils were detected. Among nine ABC transporters analyzed, MRP1 is expressed signifieandy higher in tumor tissues than in normal controls. Co-focus microscopy analysis showed that the majority of MRP1 positive cells were CD133 positive. [Conclusion] ABC transporter MRP1 is highly expressed in human eolorectal cancers comparing with normal tissues, and is primarily expressed by CD133 positive cancer cells.
