摘要
目的研究小鼠OPG基因敲除对其骨质量及微结构的影响。方法 HE、扫描电镜、四环素荧光及显微CT观察OPG基因敲除小鼠骨微结构的改变;DXA仪测定小鼠股骨标本的总体骨密度和骨矿含量。结果与野生型小鼠相比,OPG基因敲除小鼠松质骨与皮质骨均出现明显的骨丢失,伴显著的骨微结构破坏;总体骨密度和骨矿含量减少。结论 OPG-/-小鼠发生明显骨丢失,骨微结构破坏。
[Objective] To investigate the changes of bone quality and micro-architecture in OPG knockout (OPG^-/-) mice. [Methods] Bone micro-architecturc and loss of bone mass were assessed by hematoxylineosin staining, scanning electron microscope and micro-CT. BMD and BMC were detected by DXA. [Resuits] Compared with wild-type mice, OPG4^-/- mice showed a loss of BMD and disturbance of micro-architecture. The BMD and BMC were reduced by DXA determination. [Conclusions] The disturbant bone micro-architecture and lowered BMD were observed in OPG^-/- mice.
