摘要 目的 研究光性屈光性角膜切削术(PRK)后,转移生长因子β受体Ⅱ(TGFβRⅡ) 和表皮生长因子受体(EGFR)mRNA 在角膜的表达变化,探讨角膜上皮下混浊(haze) 形成的发生机制。方法 新西兰白兔16 只,随机分成4 组。其中12 只施行PRK,分别于术后1 、2、3 个月用裂隙灯显微镜观察haze 形成情况,并用原位核酸分子杂交方法,检测角膜上皮和基质TGFβRⅡ和EGFRmRNA的表达。结果 PRK后1 个月已有haze 形成,2 个月时haze 最明显,术后3 个月haze 减轻或消失。正常兔角膜上皮和基质有TGFβRⅡ和EGFRmRNA表达,PRK后角膜TGFβRⅡ和EGFRmRNA表达增加,且以术后1、2 个月表达最明显。TGFβRⅡ和EGFRmRNA表达强弱与形成haze 的轻重相关。结论 TGFβRⅡ和EGFR参与PRK后伤口愈合过程,且调节着haze Objective To study the mechanism of haze formation and investigate the expression changes of transforming growth factor β receptor Ⅱ(TGF βRⅡ) and epidermal growth factor receptor(EGFR) mRNA in corneal epithelium and stroma after photorefractive keratectomy (PRK). Methods Sixteen New Zealand white rabbits were randomly divided into 4 groups,and PRK was performed on each eye of 12 rabbits. The haze formation was examined under a split lamp microscope at the 1st, 2nd and 3rd month after PRK, and the expressions of TGF βRⅡ and EGFR mRNA were assessed with in situ hybridization. Results The cornea haze had formed at the 1st month after PRK. The most prominent haze formation was observed at the 2nd month, and decreased even disappeared at the 3rd month after PRK. TGF βRⅡ and EGFR mRNA expressions were present on the normal corneal epithelium and stroma. The capacity for expressions of TGF βRⅡ and EGFR mRNA increased after PRK, and the peak appeared on the 1st and 2nd month. The extents for expression of TGF βRⅡ and EGFR mRNA were in corelation with the haze formation.Conclusions TGF βRⅡ and EGFR take part in promoting corneal wound healing after PRK, and may contribute to cornea haze formation and development.
