摘要
目的:研究丹参对人粘连成纤维细胞增殖的抑制作用。方法:从人体腹膜粘连组织中原代培养出成纤维细胞(AFB),以地塞米松为阳性对照,考察丹参及其主要成分丹参素、丹参酮ⅡA对AFB增殖活性与生长因子-β1(TGF-β1)mRNA表达的影响。结果:丹参冻干粉、丹参素作用下AFB活性下降幅度大,而且半数抑制浓度(IC50)显著低于丹参酮ⅡA,TGF-β1mRNA表达水平亦显著降低(P<0.01或P<0.05),丹参酮ⅡA对TGF-β1表达无显著影响。结论:丹参通过下调AFBTGF-β1mRNA表达抑制其增殖,主要起效成分为丹参素。
OBJECTIVE:To study the inhibition effect of Salvia miltiorrhiza on the proliferation of human adhesive fibroblasts.METHODS:Adhesive fibroblasts(AFB) from human peritoneal adhesions were harvested by primary culture.The effect on proliferation and TGF-β1 mRNA expression of AFB by danshensu and danshinone ⅡA were observed using dexamethasone as positive control.RESULTS:The activity of AFB treated with S.miltiorrhiza freeze-dried powder and danshensu decreased significantly,and IC50 was significantly lower than danshinone ⅡA.TGF-β1 mRNA expression decreased significantly(P0.01 or P0.05).Danshinone ⅡA had no significant effect on TGF-β1 expression.CONCLUSION:S.miltiorrhiza inhibit the proliferation of AFB by down-regulating TGF-β1 mRNA expression.Main active component is danshensu.
