内皮型一氧化氮合酶基因G894T多态性与颈动脉粥样硬化的相关性 被引量:3

Study of association between G894T polymorphism of endothelial nitric oxide synthase gene and carotid atherosclerosis

摘要 目的探讨天津市汉族老年人群内皮型一氧化氮合酶(eNOS)基因G894T多态性与颈动脉粥样硬化(CAS)的关联性。方法选择接受颈动脉超声检查患者427例,根据超声检查结果分为CAS组1 30例和对照组297例。采用PCR-RFLP方法分析G894T多态性基因型,同时对所有对象检验血脂等危险因素。结果 CAS组基因型GT、GT+TT和T等位基因频率明显高于对照组(P<0.05,P<0.01)。调整了年龄和性别后,基因型GT+TT与CAS的关联差异有统计学意义(OR=1.89,95%CI:1.20~2.98,P=0.007)。对其他危险因素调整后,logistic回归分析,基因型GT+TT不是CAS的独立危险因素(OR=1.43,95%CI:0.85~2.40,P=0.1 78)。在CAS组中,不同斑块类型的分布、斑块总面积和Crouse积分在T等位基因携带者与非携带者差异无统计学意义(P>0.05)。结论 eNOS基因G894T多态性的基因型GT+TT与CAS的发生相关,但不是CAS的独立危险因素,与CAS的严重程度亦无关联。 Objective To investigate the association between endothelial nitric oxide synthase (eNOS) gene G894T polymorphism and carotid atherosclerosis(CAS) in elderly Han people living in Tianjin. Methods Four hundred and twenty seven patients undergoing B-mode ultrasonography of carotid artery were enrolled in the study. Of the total, 130 had CAS and comprised the CAS group, other 297 comprised the control group. The eNOS gene G894T polymorphism was analyzed by PCR RFLP. The plasma lipid levels and other risk factors were determined in all subjects. Resuits The frequencies of T allele and GT,GT+TT genotypes were significantly higher in CAS group than in control group (P〈0.05,P〈0.01). After adjusting age and gender, the association between GT+GG genotype and CAS was statistically significant (OR = 1.89,95%CI: 1.20-- 2.98, P = 0. 007). After adjustment for other risk factors, the logistic regression analysis found that GT+GG genotype was not an independent risk factor for CAS (OR = 1.43,95% CI: O. 85-- 2.40, P = 0. 178). In CAS group,no significant difference in the distribution of different types of plaques, the total area of plaques and the Crouse score of plaques between people carring T allele or not was observed (P) 0.05). ConcLusion The eNOS gene G894T polymorphism was associ- ated with CAS,but not its severity with statistical significance, and was not an independent risk factor for CAS.
