反相高效液相色谱技术分析种子醇溶蛋白鉴定小麦品种 被引量:1

摘要 用反相高效液相色谱技术分析了我国北部冬麦区18个不同小麦品种的种子醇溶蛋白,发现小麦种子醇溶蛋白的异质性极强,其组成随基因型的不同而有所变异。用70%乙醇直接提取的所有不同品种种子醇溶蛋白的色谱图型差异显著,且均表现出各自明显的特异性。因此,应用反相高效液相色谱技术分析小麦种子醇溶蛋白可以对小麦品种进行鉴定。 Wheat prolamin (gliadin) protein fraction extracted from 18 winter wheat varieties in north-china region was analyzed by reversed-phase high-performance liquid chromatography (RP-HPLC). Results indicated that gliadins are very heterogeneous, and their composition varies among wheat genotypes. Chromatograms are presented that demonstrate that significant qulitative and quantitative differences occur among all 18 wheat varieties examined, and each have its considerable characteristic genetic fingerprints. Therefore,wheat varieties can be readily differentiated and thereby identified through RP-HPLC of their gliadins directly extracted from wheat flour with aqueous 70% ethanol.
