摘要
目的:通过电项针干预睡眠剥夺大鼠的实验研究探讨电项针对失眠诱发氧化应激反应影响机制。方法:将大鼠随机分为空白组、模型组、电项针组、黄芪注射液组,除空白组外,其余组通过腹腔注射PCPA复制睡眠剥夺大鼠模型。通过光栅分光光度计观察脑干中SOD及NO含量的变化。结果:光栅分光光度计法证实,睡眠剥夺后,电项针组和黄芪注射液组大鼠脑干网状组织内NO、SOD的含量降低,与模型组比较均有显著差异(P<0.01);而此两组比较无显著性差异(P>0.05)。电镜下观察可见,睡眠剥夺后,模型组大鼠脑干细胞膜边界不清晰,细胞浑浊,细胞内有空泡变性,线粒体空泡变性,肿胀明显粗面内质网含量减少,突触融合;而电项针组未见明显变化。结论:电项针可通过减轻睡眠剥夺大鼠的氧化应激反应,减少睡眠剥夺大鼠神经毒性作用。
Objective:To discuss the mechanism of the electro-nape-acupuncture on the effect of rats of sleep deprivation and oxidative stress induced by it through the intervention of sleep deprivation experiments on rats.Methods:Rats were randomly divided into control group,model group,the acupuncture group,and astragalus injection group.In addition to the control group,other groups were duplicated of sleep deprivation rats models by intraperitoneal injection of PCPA.Observe the content of NO and SOD by grating spectrophotometer method.Results:Confirmed by grating spectrophotometer method,the electro-nape-acupuncture and astragalus injection could decrease the contents of NO and SOD in the brainstem reticular of the sleep deprivation rats.Compared with the model group,there were significant differences(P0.01).Compared with one another,the difference was no significant(P0.05).Observe by electron microscopy,in the model group the borders of cells in brain stem of rats was not clear,mitochondria degenerated clearly,the contents of rough endoplasmic reticulum decreased,synaptic integrated,the cells of rats in the electro-nape-acupuncture group did not change significantly.Conclusion:Electro-nape-acupuncture can reduce the oxidative stress response,and neurotoxicity can be decreased among sleep deprivation rats through electro-nape-acupuncture..
