摘要
[目的]探讨乌梅丸对大鼠溃疡性结肠炎(UC)结肠组织白细胞介素8(IL-8)及白细胞介素13(IL-13)表达的影响。[方法]采用2,4-二硝基氯苯(DNCB)加醋酸复合法制备大鼠UC模型。动物随机分为正常对照组、模型组、柳氮磺胺吡啶(SASP)组和乌梅丸大、中、小剂量组6组。采用免疫组织化学染色法检测结肠组织IL-8、IL-13的表达。[结果]与正常组相比,模型组大鼠结肠组织中IL-8的阳性表达显著增高(P<0.01),而乌梅丸各剂量组、SASP组则较模型组明显降低(P<0.01)。与正常组相比,模型组大鼠结肠组织中IL-13的阳性表达显著降低(P<0.01),而乌梅丸各剂量组、SASP组则较模型组明显升高(P<0.01)。[结论]IL-8I、L-13参与了UC的发生。乌梅丸可能是通过下调IL-8,上调IL-13达到治疗UC的目的。
[Objective]To investigate the impact of Wumei pills on the expression of the interleukin-8 and interleukin-13 in the colon tissue of rats with ulcerative colitis(UC).[Methods]2,4-Dinitrochlorobenzene(DNCB)plus acetate was used to prepare rat model of UC.Animals were randomly divided into six groups including control,DNCB group,salicylazosulfapyridine(SASP)group and Wumei pill high dose,medium dose and low dose group.The expression of interleukin-8 and interleukin-13 in the intestinal tissue were tested by immunohistochemical staining.[Results]The expression of interleukin-8 in the colon tissue was significantly increased in DNCB group compared with that in control group(P0.01),and that in Wumei pill large dose,medium dose,small dose group or SASP group was significantly lower than that in the model group(P0.01).The level of interleukin-13 expression in the colon was significantly lower in the model group than in normal group(P0.01),and that in Wumei pill hign dose,medium dose,low dose group or SASP group was significantly higher than that in the model group(P0.01).[Conclusion]Interleukin-8 and interleukin-13 are involved in the pathogenesis of UC.Wumei pill has been shown to inhibit the expression of interleukin-8 and increase the expression of interleukin-13,which provide an alternative drug in the treatment of UC.
