摘要 的:建立高效液相色谱法测定人血清中西替利嗪。方法:用HPLC法检测8名健康自愿者血中西替利嗪浓度,经3P87程序拟合,计算药动学参数。结果:西替利嗪药动学参数T1/2α,T1/2β,Tmax,Cmax,V/F和AUC分别为(1.6±0.8)h,(7.2±1.1)h,(1.3±0.4)h,(374.7±61.8)ng·ml-1,(39.1±8.8)L和(3188.4±246.1)ng·h·ml-1。结论:该方法简单、快速、准确,适用于药动学及药效学研究。 OBJECTIVE:To establish an HPLC method for determination of cetirizine in human serum.METHODS:Cetirizine concentration in serum of healthy male volunteers was determined with HPLC,and the pharmacokinetic parameters was simulated with 3P87 program.RESULTS:The pharmacokinetic parameters T 1/2α , T 1/2β , T max , C max , V/F , and AUC were( 1.6 ± 0.8 ) h,( 7.2 ± 1.1 ) h, ( 1.3 ± 0.4 ) h, ( 374.7 ± 61.8 ) ng·ml -1 ,( 39.1 ± 8.8 ) L,and ( 3 188.4 ± 246.1 ) ng·h·ml -1 ,respectively.CONCLUSION:This method is simple,rapid,and accurate.It is useful for research on pharmacodynamics and pharmacokinetics of cetirizine.
