摘要 目的:探讨碱性成纤维细胞生长因子(bFGF)反义寡核苷酸对胶质瘤细胞凋亡的诱导作用。方法:将硫代磷酸修饰的bFGF反义寡核苷酸转染人脑胶质瘤细胞系TJ905,采用原位细胞死亡检测方法和电子显微镜,观察胶质瘤细胞凋亡的形态学变化,并用免疫组化法检测了增殖细胞核抗原(PCNA)的变化。结果:胶质瘤细胞呈现凋亡的形态改变,PCNA的表达明显降低。结论:bFGF反义寡核苷酸可诱导胶质瘤细胞的凋亡,抑制胶质瘤细胞的增殖。bFGF基因可作为反义治疗的目的基因。 Aim: To test the hypothesis that bFGF antisense oligodeoxynucleotides may induce the apoptosis of glioma cells. Methods: bFGF antisense oligodeoxynucleotides were transfected to TJ905 human glioma cells. The morphological changes of the cell were evaluated by TdT-mediated dUTP nick end labeling and electron microscope and PCNA by immunohistochemical staining. Results: The glioma cells transfected with bFGF antisense oligodeoxynucleotides displayed apoptotic morphological changes and the expression level of PCNA decreased dramatically. Conclusion: bFGF antisense oligodeoxynucleotides can induce the apoptosis of human glioma cells and inhibit their proliferation. bFGF gene could be chosen as target gene in the anti-sense gene therapy for human gliomas.
