摘要
目的 阐示胰腺良恶性疾患细胞凋亡的存在形式及凋亡介导基因ICE在胰腺良恶性疾患中的表达水平。方法 由凋亡原位分析、免疫组化及Western 印迹杂交,对38 例胰腺癌石蜡标本、21 例胰腺癌冰冻标本、20 例慢性胰腺炎冰冻标本进行凋亡及ICE表达的半定量统计学分析。结果 肿瘤细胞凋亡原位分析结果为1-55±0-89,同组织非肿瘤细胞为2-34±0-71,二者差异有极显著意义(P<0-01) ;免疫组化示ICE在胰腺癌组织中的表达为1-33±0-79,慢性胰腺炎组织为2-60 ±0-81,二者差异有极显著意义(P<0-01);Western 印迹示ICE前体P45 及中间聚合体P30 在胰腺癌组织中的阳性率为9/21(42-9% ),慢性胰腺炎组织为16/20(80 %) ,二者差异有极显著意义(P<0-01)。结论 肿瘤细胞凋亡状态的减弱及ICE在胰腺良恶性疾患间的表达差异提示正常状态凋亡机制的被抑制是细胞癌变的可能途径。
Object The presence of apoptosis and the expression of ICE, a kind of apoptosis mediating gene, in tissues of human pancreatic carcinoma and chronic pancreatitis were investigated. Method Thirty eight paraffin embedded tissues of pancreatic carcinoma were used for in situ apoptosis assay, and 21 frozen tissues of pancreatic carcinoma and 20 frozen tissues of chronic pancreatitis were used for both immunohistochemistry and Western blot analyses. In addition, the staining intensities observed from in situ apoptosis assay and immunohistochemistry were scored semiquantitatively. Results With regard to in situ apoptosis assay, the staining score for tumor cells (1.55±0.89) was significantly lower than that of benign cells (2.34±0.71) within the same sections (P<0.01). Similarly, the immunohistochemistry staining with the antibody against ICE in the carcinoma tissues showed significant difference in comparison with that in the chronic pancreatitis tissues (1.33±0.79 vs 2.60±0.81, P<0.01). Western blot analysis revealed that the positive bands migrating at Mr. 45?000 and Mr. 30?000 were detected in 9/21 (42.9%) of the carcinoma tissues, whereas in 16/20(80%) of the chronic pancreatic tissues. There was also significant difference (P<0.01) between the two groups. Conclusion The suppressed apoptotic state of the tumor cells and the expression difference of ICE in malignant and benign pancreatic tissues indicate that the inhibition of normal apoptosis is a possible way for carcinogenesis.
