摘要
目的探讨鞘内注射芬太尼对急性炎性内脏痛大鼠背根神经节(DRG)电压门控Nav1.8的影响。方法将24只SD大鼠,用随机数字表法分为3组,每组8只,腹腔注射乙酸建立急性炎性内脏痛模型,实验中经L3、L4的留置管给药。对大鼠进行行为学评分,灌注取材L5或L6DRG,用免疫荧光染色方法观察Nav1.8表达的变化。结果单纯乙酸组行为学评分高于芬太尼组(P<0.05);与空白对照组相比,单纯乙酸组、芬太尼组Nav1.8表达均增加,而芬太尼组Nav1.8表达程度低于单纯乙酸组(P<0.05)。结论鞘内注射芬太尼对急性炎性内脏痛大鼠的镇痛效应不仅与阿片受体有关,与Nav1.8也关系密切。
Objective To explore the effects of intrathecal injection of fentanyl on the expression of NaV1.8 in dorsal horn from the rats modeled in visceral pain of acute inflammatory reaction.Methods 24 male SD rats were randomly divided into 3 groups by means of random number table(n=8):group Ⅰ blank control group Ⅱ and Ⅲ were exposed to acetic acid,fentanyl respectively.The intraspinal intubation of L3-4 level to those rats and the visceral pain model of an acute inflammatory reaction was made by injecting acetic acid into peritoneal cavity.Then the behavior scores were marked.After the observation stage,rats in each group were respectively perfused with paraformaldehyde and the materials were taken from L5 or L6 dorsal root ganglions,and the changes of expression of Nav1.8 were observed with immunofluorescence staining.Results The ethological grade of group acetic acid,was higher than that of the fentanyl group(P0.05),while compared with the group blank controls,the expression of Nav1.8 increased in both groups,but the increasing range of the Nav1.8 expression in fentanyl group was lower than that of acetic acid group.Conclusion Intrathecal injection of fentanyl has the analgesic effect on those rats which modeled in an acute inflammatory visceral pain.It not only relates to the opium receptor,but also has substantial connection with Nav1.8.
