芪蓝颗粒干预大鼠舌黏膜癌变的Survivin基因表达的研究 被引量:1

摘要 目的:观察芪蓝颗粒对4-硝基喹啉-1-氧化物(4NQO)诱导的大鼠舌癌变的抑制及对生存素基因的影响。方法:SD大鼠150只,随机分为癌变模型组,低、中、高剂量中药干预组以及正常对照组,以4NQO饮水喂养诱导大鼠舌癌变,同时给予芪蓝颗粒干预癌变过程,取9、18、27、36周舌标本进行组织病理学观察,采用逆转录-聚合酶链反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)方法检测这150例舌组织中生存素基因的表达。结果:27、36周时,不同组癌变率差异明显(P<0.05),B、C、D组的癌变率均低于A组,C组的癌变率最低。在舌异常增生和癌组织中,中药干预组中生存素基因的表达强度明显低于A组(P<0.05)。结论:芪蓝颗粒有确切的抑制口腔癌变功效,对细胞凋亡的调控可能是其抑癌机制基础之一。 Objective: To examine the inhibitory effect of Chinese herb Qilan granulates and its effect on expression of Survivin on 4-nitro-quinoline 1-oxide(4NQO)-induced tongue carcinogenesis in rats.Methods: A total of 150 SD rats were divided into five groups: carcinogenesis model group,three administration subgroups and normal control group.Rats in group A,B,C,D were fed with 0.002% 4NQO dissolved in drinking water to induce tongue carcinogenesis in rats.Different concentration of the herb Qilan granulates was given to the rats in group B,C,D during oral carcinogenesis.Rats were sacrificed at 9,18,27and 36 weeks respectively from the beginning of the experiment.The tongues of the rats were dissected for gross and histological assessment,and reverse transcription-polymerase chain reaction(RT-PCR) was also employed to detect the expression of Survivin in the specimens.Results: Compared with the model group,the incidence of dysplasia after 27 and 36 weeks and the expression of Survivin in tongue dysplasia and carcinomas tissue were suppressed by Qilan granulates(P0.05).Conclusion: Administration of Qilan granulates effectively suppressed 4NQO-induced tongue carcinogenesis.It may exert the chemopreventive effects by the regulation of cell apoptosis.
