TRIP3基因与儿童急性白血病免疫分型关系的研究

Correlation between the expression of TRIP3 gene and immunological classification in children with acute leukemia

目的研究TRIP3(thyroid hormone receptor interacting protein 3)基因在儿童急性白血病中的表达,探讨其与儿童急性白血病免疫分型关系。方法采用逆转录-聚合酶链反应法(RT-PCR)及流式细胞仪检测检测39例儿童急性白血病的免疫表型及TRIP3基因水平,并分析二者的关系。结果 TRIP3基因在儿童急性淋巴细胞白血病与儿童急性非淋巴细胞白血病中的表达水平无明显差异(P>0.05);TRIP3在不同类型儿童急性淋巴细胞白血病中表达情况不同,31例初治患者,B-ALL21例,TRIP3阳性13例,阳性率为61.9%,T-ALL7例,TRIP3阳性2例,阳性率为28.5%,二者差异显著(P<0.05),且B-ALL TRIP3表达水平高于T-ALL(P<0.05)。结论 TRIP3基因在B系淋巴细胞白血病表达水平高,检测TRIP3基因可以估计急性淋巴细胞白血病的临床预后。

Objectives To study the expression of thyroid hormone receptor interacting protein 3（TRIP3） gene in acute leukemia of children,and discuss its relation with acute leukemia of children on immunological classification.Methods The immunological expression of acute leukemia of children and genes of TRIP3 on 39 cases were detected by RT-PCR and flow cytometry,and their relationship was analyzed.Results There was no significant difference in expression of TRIP3 gene between acute lymphoblastic leukemia and acute myelogenous leukemia of children;there was certain relevance between the genes expression and immunological classification of acute leukemia with 13 positive TRIP3 cases,which was 61.9% of 21 cases of B-ALL and 2 positive TRIP3 cases,which was 28.5% of 7 cases of T-ALL among 31 patients being treated.The TRIP3 expression of B-ALL was higher than that of T-ALL（P0.05）.Conclusion The TRIP3 gene expresses highly in the B-ALL acute leukemia of children,the ratio of TRIP3 osoform may act as a useful marker to evaluate the clinical prognosis.
