贵州省首次从山羊分离到布鲁氏菌及其种型鉴定 被引量:27

摘要 目的从贵州布鲁氏菌抗体阳性的山羊血液分离布鲁氏菌病原体并对其进行种型鉴定。方法采用血培养法对经试管凝集试验检测为布鲁氏菌抗体阳性的山羊血血液进行细菌分离培养,并应用传统方法和和分子生物学方法对可疑菌落进行种型鉴定。结果 11份(GZB 01-11)抗体阳性的山羊血标本中有4份(GZB03、GZB04、GZB09、GZB11)经血培养瓶培养出布鲁氏菌可疑菌落,可疑菌落经传统方法鉴定为羊种生物3型布鲁氏菌,4株菌进一步采用布鲁氏菌属特异性PCR(BCSP31-PCR)鉴定为布鲁氏菌属细菌,GZB03,GZB04,GZB11经种/型特异性PCR(AMOS-PCR)将其定为羊种布鲁氏菌,而GZB09采用该方法不能定种。结论从11份贵州省布鲁氏菌抗体阳性山羊血样中分离到4株羊种布鲁氏菌,首次证实贵州省存在羊种布鲁氏菌。 To isolate Brucella strains from the blood of Brucella antibody positive goats in Guizhou province and identify the isolated suspicious strains with conventional and molecular techniques, blood culture method was used to isolate Brucella strains from the blood of anti-Brucella antibody positive goats detected with standard agglutination test (SAT). Conventional and molecular techniques were used to identify suspicious bacteria strains. The results showed that Brueella suspicious colonies were cultured in 4 (GZB03, GZB04, GZB09, GZBll) of the total 11 (GZB01-11) goat blood samples. 4 strains were identified as B. melitensis biotype 3 by conventional tests and were further identified as Brucella spp. by genus specific BCSP31-PCR. Strain GZB0a, GZB04, and GZBll were classified as B. melitensis by species-specific AMOS-PCR, while GZB09 was unidentifiable to species. It is the first time to confirm that B. rnelitensis exists in Guizhou province.
