摘要
为了解我国南方某城市儿童尿液中单羟基菲的含量,于2007年11月采集该市一幼儿园80名4-6岁儿童尿样。尿液经过酶解、固相萃取(SPE)富集、净化,采用高效液相色谱-荧光检测器检测,共检测出4种单羟基菲,且以9-羟基菲为主。除4-羟基菲的检出率为53.75%外,其余单羟基菲的检出率近乎100%。各单羟基菲的中位数分别为(μmol/mol肌酐):2-+3-羟基菲(0.13)、9-羟基菲(1.64)和4-羟基菲(0.06)。研究人群尿中总单羟基菲含量的中位数高于该省南部沿海中学生,与该省北部多环芳烃高暴露环境背景人群相当,显著高于美国类似人群。9-羟基菲与总单羟基菲的浓度相关性显著,r值为0.979,显示9-羟基菲可以作为生物标志物,用于指示人体尿中单羟基菲的浓度水平。
To analyze the concentration of hydroxyphenanthrene in urine of kindergarten infants,80 children(4-6 years) urine samples from a kindergarten in a city in South China were collected in November 2007.After enzymic hydrolysis,the urine samples were preconcentrated and cleaned up by solid phase extraction,and then the urinary hydroxy-PAHs(HOPAHs)were determined using high performance liquid chromatography with fluorescence detection.Four kinds of hydroxyphenanthrenes were detected in urinary samples,with detection frequencies of 4-hydroxy phenanthrene as 53.75%, and the rest close to 100%.The median concentrations of those hydroxyphenanthrenes were(μmol/mol creatinine)2-+3hydroxyphenanthrene(0.13),9-hydroxyphenanthrene(1.64)and 4-hydroxyphenanthrene(0.06).The median concentration of those∑hydroxyphenanthrenes was equal to people where near emission source of PAHs in north and higher than those of the students in South of the Province,but higher than the reported data from similar background population in USA.There were relatively good correlations with correlation coefficient of 0.979 between 9-hydroxyphenanthrene and∑hydroxyphenanthrenes, indicating that 9-hydroxyphenanthrene may be a biomarker to represent the level of hydroxyphenanthrenes in human urine.
