GCDCA对肝癌SMMC7721细胞诱导分化的影响 被引量:1

摘要 目的探讨甘氨鹅脱氧胆酸(glycodeoxycholicacid,GCDCA)对人肝癌细胞SMMC7721诱导分化的影响。方法采用96孔细胞培养板体外培养人肝癌细胞株SMMC7721,GCDCA分别处理细胞48,72,96h,采用放射免疫法检测肝癌sMMC7721细胞的甲胎蛋白(AFP)、清蛋白(ALB)分泌量,同时利用免疫组化分析肝癌SMMC7721细胞AFP表达的影响。结果GCDCA能显著抑制肝癌细胞SMMC7721AFP的分泌,降低其表达,且能大大提高ALB的分泌。其效果GCI)-CA200umol/L处理组优于GCDCA100umol/L处理组,均有时效和量效的关系。结论GCDCA能抑制肝癌SMMC7721细胞AFP的表达和分泌,提高ALB的分泌,具有诱导肝癌细胞SMMC7721细胞良性分化的能力。 Objective To investigate the effect of differentiation-inducing of glycodeoxycholic acid(GCDCA) on human hepatocellular carcinoma SMMC7721 cells. Methods SMMC7721 cell were cultured in 96 cell culture plate and then the cells were treated by 100 and 200 umol/L GCDCA groups respectively at 24,48 and 72 h. The content of AFP,ALB of the culture supernatant was determined by immunoradiometric assay. Simultaneously,the AFP expression of SMMC7721 cells treated by GCDCA was detected by immunohistochemical staining. Results The AFP secretes of SMMC7721 cells was obviously inhibited and the content of ALB was increased with the time-dosage dependent relationship. And then the AFP expression of SMMC7721 cells was reduced apparently by GCDCA and the effects of GCDCA 200 umol/L groups was better than the GCDCA 100 umol/L groups. Conclusion GCDCA could inhibit the secret and expression of AFP. And then increase the secret of ALB. Therefore GCDCA could induce the SMMC7721 cells differentiation.
