摘要
BYSL基因是哺乳动物胚胎着床和胚胎发育的关键因子。为进一步研究猪BYSL基因的结构与功能,揭示其表达模式,本研究结合基因同源序列克隆技术和RT-PCR方法克隆了猪BYSL基因;利用生物信息学软件对该基因进行序列特征和结构分析;利用Real-time PCR技术分析了BYSL基因的时空表达模式。结果表明,猪的BYSL编码区全长为1 308 bp,共编码435个氨基酸;猪BYSL含有一个Bystin结构域;BYSL蛋白序列与人(90%)和牛(90%)BYSL的蛋白序列具有很高的相似度。组织表达模式研究发现,猪BYSL基因在卵巢和子宫组织中均有较高水平的表达;子宫组织发育表达模式研究发现,猪BYSL基因在性成熟前表达量逐渐升高,4月龄达到最大值。研究发现BYSL基因在哺乳动物中具有相似的生物学功能,猪BYSL基因在繁殖过程中也可能发挥重要作用。
BYSL gene plays an important role during the implantation.In order to further elucidate the relationship between the structure and function of BYSL,the porcine BYSL(pBYSL) gene was isolated using bioinformatics and RT-PCR methods,sequence analyses were done using boinformatics tools,and expression profile was analyzed by Real-time PCR.Our results suggested that the coding sequence of pBYSL was 1 308 bp,encoding 435 amino acids which including a bystin domain.pBYSL exhibited high homology to human(90%) and bovine(90%) Real-time PCR analyses showed that pBYSL was expressed in both ovary and uterus.Before sex mature,the expression level gradually decreased and reached the top in 4-month old.The BYSL gene was highly conservative during evolution.This research suggested that BYSL had approximated biological function among mammalian,so pBYSL might play a role during porcine reproduction.
