乌苏里江唇(Hemibarbus labeo Pallas)的细胞遗传学分析

Cytogenetic analysis of Hemibarbus labeo Pallas

采用体内注射PHA和秋水仙素、肾细胞短期培养、常规空气干燥方法,制备乌苏里江唇的染色体。对其肾细胞染色体数目统计分析结果表明,乌苏里江唇的染色体数染色体组有50条染色体,8对中部着丝点染色体(m),8对亚中部着丝点染色体(sm),9对端部着丝点染色体和亚端部着丝点染色体(st,t),其染色体臂数(NF)为82。乌苏里江唇组型公式为2n=16m+16sm+18st+t。采用流式细胞分析仪,测定了乌苏里江唇的DNA含量,与鸡血细胞标准对照相比为1.02±0.10,以鸡红细胞DNA含量2.3 pg.N-1计,则乌苏里江唇的体细胞DNA含量为2.34 pg.N-1。与鲤形目其他科的二倍体鱼类比较,乌苏里江唇染色体数目和DNA含量相互对应,共同体现出二倍体特征。

Metaphase chromosome specimens of Hemibarbus labeo injected with PHA and colchicin,were prepared from short-term culture of kidney cells with air-drying technique.Its diploid chromosome number was 2n=50.The karyotype is composed of 16m,16sm and 18st＋t chromosomes,NF=82.Diploid nucleus DNA content was measured from the somatic cells of Hemibarbus labeo pallas using flow cytometer（Beckman cellL ab Quant,made in USA） and the DNA content（2.3 pg·N-1） oferythrocytes of chick（Gallus sp） as standard.The Hemibarbus labeo Pallas DNA content was 2.34 pg·N-1,which was correspond with the result measured chromosome number as ploidy.According to the present results and date reported,Hemibarbus labeo Pallas was possibly a diploid（2n）.
