PARP1在肿瘤中的作用研究

摘要 PARP1是真核细胞内具有多聚腺苷酸二磷酸核糖基(PAR)催化活性的蛋白酶,在DNA损伤断裂时被激活,参与DNA的修复,在DNA修复和细胞凋亡中发挥至关重要的作用。PARP1的缺失能够使DNA对细胞损伤易感,从而诱导肿瘤的发生。目前的研究结果显示PARP1在肿瘤细胞中的过高表达提示在临床中运用PARP1抑制剂来抑制PARP1在DNA损伤后修复的活性,从而抑制PARP1参与介导的细胞DNA损伤修复,对肿瘤有治疗有一定的效果。本文就PARP1的结构与功能,PARP1在肿瘤的发生发展中的作用以及PARP1与肿瘤治疗的关系进行总结和综述。
