摘要
采用实时定量PCR技术对类猪圆环病毒因子P1感染猪不同时相猪肺泡巨噬细胞中外源性抗原加工递呈相关分子SLA-DR、SLA-DM和CD74及共刺激分子CD40、CD80和CD86 mRNA的表达进行了检测。结果表明,病毒感染后3 d,上述分子的mRNA表达皆低于对照组相应分子的mRNA表达。其中,CD80 mRNA的表达呈下调趋势(P<0.1),CD40、CD74和SLA-DR的mRNA的表达显著下调(P<0.05);感染后8 d的SLA-DR和SLA-DM以及感染后17 d SLA-DM的mRNA的表达有下调趋势(P<0.1);感染后24 d的CD86 mRNA的表达呈上调趋势(P<0.1);感染后40 d的CD74 mRNA的表达显著下调(P<0.05)。结果揭示,类猪圆环病毒因子P1感染对猪肺泡巨噬细胞的外源性抗原加工和递呈功能有显著影响,导致其外源性抗原递呈功能下降及紊乱,进而使机体的体液免疫应答功能受到影响。
The transcriptional levels of SLA-DR,SLA-DM,CD74 and costimulatory molecules CD40,CD80 and CD86 of porcine pulmonary alveolar macrophages(PAMs) on different phases after porcine circovirus-like agent P1 infection were detected by real-time RT-PCR.The results showed that mRNA levels of all above-mentioned molecules were down-regulated at 3 days post-inoculation(DPI).Among those CD80 mRNA expression showed a downward trend(P0.1),and CD40,CD74 and SLA-DR mRNA expression were significantly declined(P0.05).The mRNA expression of SLA-DR SLA-DM at 8 DPI and SLA-DM at 17 DPI were down-regulated(P0.1).CD74 mRNA expression decreased markedly at 40 DPI(P0.05).Whereas the expression levels of CD86 at 24 DPI showed an upward trend(P0.1).These results suggest that P1 infection affected markedly processing and presentation of PAMs for exogenous antigen,which resulting in a decrease and unbalance of antigen presentation function of PAMs for exogenous antigen and impairment for humoral immune responses.
