槲寄生碱体外通过激活Caspase-3诱导涎腺腺样囊性癌细胞凋亡 被引量:1

摘要 目的证实槲寄生碱(mistletoe alkali,MA)通过激活Caspase-3诱导人涎腺腺样囊性癌细胞(ACC)凋亡。方法将ACC细胞分为三组,即对照组、2.24μg/ml槲寄生碱处理组和1.12μg/ml槲寄生碱处理组,采用荧光染色法和免疫组织化学染色方法,检测三组细胞中Caspase-3蛋白的表达情况。结果槲寄生碱组Caspase-3表达明显高于对照组(P<0.01),并且药浓度越高,Caspase-3表达越强。结论槲寄生碱可通过激活Caspase 3诱导ACC细胞凋亡,并呈剂量依赖性。 Objective To study whether alkaloid from mistletoe induces apoptosis of cells of adenoid cystic carcinoma of salivary gland(ACC cell) via activation of caspase3.Methods the cells were divided into three groups(control group 2.24 μg/ml alkaloid group,1.12 μg/ml alkaloid group),the expressive status of caspase3 in three groups were detected by fluorescence and immunohistochemistry staining,the expression of alkaloid groups is obviously higher than the control groups.Results The expression of caspase3 of alkaloid group was much higher than that of control groups(P0.01),and the expression is stronger with the higher concentration of alkaloid.Conclusion alkaloid of mistletoe induces apoptosis of ACC cell via activation of caspase3 and the increased caspase3 activity is dose dependent.
