摘要
[目的]从加味茵芍散对TGF-β/Smad信号通路影响的角度探讨该方对肝纤维化病变的作用。[方法]按0.1 ml/kg的用量腹腔注射CCl4诱导复制家兔肝纤维化模型,50只家兔随机分成模型对照组,秋水仙碱组,加味茵芍散高、中、低剂量组5组,每组10只,通过肝脏病理取材确定肝纤维化病变,同时采用实时定量逆转录-聚合酶链反应法检测TGF-β1mRNA、Smad3mRNA、Smad7mRNA等指标。[结果]与模型组相比,秋水仙碱组和加味茵芍散高、中、低剂量组家兔肝组织TGF-β1mRNA表达明显减弱,差异有统计学意义(P<0.05);加味茵芍散各剂量组、秋水仙碱组与模型组相比家兔肝组织Smad3mRNA的表达量显著性降低(P<0.05);加味茵芍散各剂量组、秋水仙碱组与模型组相比,Smad7mRNA的表达量显著升高(P<0.05)。[结论]加味茵芍散能减轻肝细胞变性,减少胶原的合成与分泌,减轻纤维组织增生,可以下调TGF-β1mRNA、Smad3mRNA的表达,提高Smad7 mRNA的表达。该方对CCl4诱导家兔肝纤维化模型具有一定保护作用,可以延缓肝纤维化的进展,其作用机制可能与调节TGF-β/Smad信号通路有关。
[Objective] To study the influence and effect on TGF-β/Smad signal pathway in rabbit with liver hepatic fibrosis treated by JiaWeiYinShao Powder.[Methods]Liver fibrosis model in rabbits were induced and duplicated with CCl4 0.1 ml/kg by intraperitoneal injection,50 rabbits were randomly divided into model control group,colchicine group,JiaWeiYinShao powder high,medium and low dose group with 10 rabbits in each group,through the liver pathology test to determine the liver fibrosis model,while TGF-β1 mRNA,Smad3 mRNA,Smad7 mRNA and other indicators were detected by real-time quantitative reverse transcription polymerase chain reaction method.[Results] Compared with the model group,the rabbit liver TGF-β1 mRNA expression was significantly decreased in the colchicine group and the high,medium and low dose groups of JiaweiYinShao powder,the difference was statistically significant(P0.05).Compared with model group,Smad3 mRNA expression in rabbit liver tissue was significantly decreased in JiaweiYinShao powder each dose group and the colchicine group(P0.05).The expression of Smad7 mRNA was significantly higher in each dose group of JiaweiYinShao powder and the colchicine group than the model group(P0.05).[Conclusion]JiaweiYinShao powder can reduce the degeneration of liver cells,reducing the synthesis and secretion of collagen to reduce liver fibrosis.Also can reduce the expression of TGF-β1 mRNA and Smad3 mRNA while increase the expression of Smad7 mRNA.The formula has a protective effect on liver fibrosis model in rabbits induced by CCl4 and delay the progress of liver fibrosis.The mechanism may be related to the regulation of TGF-β/Smad signal pathway.
