正交实验法优化胡萝卜素的提取工艺 被引量:5

摘要 通过对胡萝卜中胡萝卜素提取最佳条件的选择,测定胡萝卜中胡萝卜素的含量。采用L(934)正交试验,以料液比、提取温度、提取时间、提取剂浓度为影响因素,根据胡萝卜素的消光系数,在492 nm计算胡萝卜中胡萝卜素含量。确定了胡萝卜中胡萝卜素的最佳提取工艺是:提取剂乙醇浓度为45%,提取温度为65℃,提取时间为55 min,料液比为1∶17(g/mL)。以胡萝卜素提取液的吸光度值及提取量为考察指标,经验证性实验得:胡萝卜素提取液吸光度值为0.771,提取量为81.92μg/mL。 Choosing the best conditions of extracting the total carotene in carrot and testing the content of total carotene.Using orthogonal design L9(34)to choose the best condition of the ratio of material and solvent,extraction temperature,extraction time and concentration of alcohol,and testing total carotene unnder 492 nm by Molar extinction coefficient of carotene.The optimum conditions for extracting Carotene from Carrot are as follows:extraction solvent is 45 % of alcohol,extraction temperature is at 65 ℃,extraction time is 55 min,the ratio of solid to liquid is 1∶17(g/mL).With the absorbency value of carotene and extraction quantity as the inspection index to do the verification experiment: carotene extract absorbance of the value of 0.771,extract amount for 81.92 μg/mL.
