盐胁迫对离体百合不定芽诱导及生理活性的影响 被引量:5

摘要 采用组织培养法研究了5种NaCl浓度0%,0.3%,0.5%,0.8%,1.2%对百合不定芽诱导、生长及丙二醛(MDA)、脯氨酸、蛋白质含量和过氧化物酶活性的影响。结果表明:百合对盐胁迫极为敏感,随NaCl浓度的增加,百合不定芽形成频率逐渐降低,不定芽及不定根生长缓慢,且在1.2%NaCl浓度下,无不定芽及根的形成。进一步的生理活性检测结果表明,百合不定芽的丙二醛、脯氨酸含量均随NaCl浓度的升高而增加,并在0.8%NaCl浓度下达到最高,分别为201.23μmol/g和225.7μg/g;蛋白质含量呈先降后增的趋势,在0.3%NaCl浓度下最低为0.96 mg/g;POD活性也随NaCl浓度的升高而增加,于0.8%浓度下达到最高,为181.60 U/(min.g)。综合盐胁迫对百合不定芽生长及生理活性的影响,0.8%NaCl浓度为百合外植体、不定芽忍耐盐胁迫的阈值。 The experiment study the effect of different NaCl concentration(0,0.3%,0.5%,0.8%,1.2%)on the adventitious bud induction,growth and its content of MDA,Pro,protein and activity of POD.The results showed that lily was very sensitive to salt stress.The formation frequency of adventitious bud was low and the growth of adventitious bud and root were slow with increase of NaCl concentration.There were no adventitious bud and root at NaCl concentration of 1.2%.Moreover,the result of physiological activity test showed that the content of MDA and Pro were increase by the increase of salt concentration and finally reached the highest content(201.23 μmol/g and 225.7 μg/g)at the salt concentration of 0.8%,The content of protein decreased to the bottom of 0.96 mg/g at 0.3% NaCl.The activity of POD reached the highest point of 181.60 U/min·g at the salt concentration of 0.8%.The Nacl concentration of 0.8% was the salt stress threshold value to explants and adventitious bud of lily.
