Effect of Ag85A Combined with GM-CSF DNA Vaccine on the Cellular Immune Function of Mice Bearing Orthotopic Bladder Cancer

摘要 【目的】探讨联合应用Ag85A与粒细胞一巨噬细胞集落刺激因子(GM—CSF)DNA疫苗对原位膀胱癌小鼠的细胞免疫的影响。【方法】以N-甲基亚硝基脲(MNU)灌注小鼠膀胱,建立小鼠原位膀胱癌模型。50只荷瘤小鼠随机分为5组:生理盐水组、空载体组、A。g85A组、联合应用组和BCG组。模型确定后d,、d。t天和dzt各肌注1次,末次注射后d,检测各组小鼠脾脏的cD4’和cD8’T细胞亚群数量及cD4。。/CD8’比值和血清7一干扰素(IFN一7)。【结果】单独应用Ag85A和联合应用Ag85A与GM-CSFDNA疫苗均能够升高CD4^+和CD8^+T细胞的百分比及CD4^+和CD8^+T的比值,提高血清IFN-7,差异具有统计学意义(P〈0.05)。联合应用的效果明显优于单独应用,差异具有统计学意义(P〈0.05),但尚不及单纯应用BCG的免疫治疗效果。【结论】联合应用Ag85A与GM—CSF DNA疫苗能够提高荷瘤小鼠的细胞免疫功能,其效果明显优于Ag85 ADNA疫苗。 [Objective] To explore the effect of Ag85A combined with GM-CSF DNA vaccine on the cellular immuni- ty in mice bearing orthotopic bladder cancer. [Methods] The mouse model of orthotopic bladder cancer was established with N-methyl-nitroso-urea(MNU) infused into mouse bladder. Fifty tumor-bearing mice were randomly divided randomly into normal saline(NS) group, pcDNA3.1 group, Ag85A group, Ag85A combined with GM-CSF group and BCG group. After the establishment of the models, the intramuscular injection was taken one time at 7th, 14th and 21th. The number of CD4^+ and CD8^+ T cell subsets, the ratio of CD4^+ / CD8^+ and serum IFN-7 in mouse spleen of each group were detected 7 days after the last injection. [Results] Both single Ag85A and Ag85A combined with GM-CSF vaccine could elevated the percentage of CD4^+ and CD8^+ T cell subsets, the ratio of CD4^+/CD8^+ and serum IFN-7 level, and there was significant difference( P d0.05). The immunotherapy effect of Ag85A combined with GM-CSF vaccine was much better than that of Ag85A vaccine alone, and there was significant difference between them( P d0.05). But the immunotherapy effect of Ag85A combined with GM-CSF vaccine was still inferior to BCG vaccine. [Conclusion]Ag85A combined with GM-CSF vaccine can increase the cellular immunity in bearing bladder tumor-bearing mice, and its effect is much better than that of Au85A DNA vaccine alone.
