摘要 目的通过体外及体内干预实验研究探讨COX-2信号通路对皮肤伤口炎症细胞、新生血管的影响,评价皮肤伤口愈合的情况,为减少伤口瘢痕形成、促进愈合的诊疗方法提供实验依据。方法采用体外细胞培养划痕试验及小鼠体内实验,运用COX-2选择性抑制剂尼美舒利进行干预,通过伤口皮肤铺片、免疫组化对伤口生长过程中炎症细胞、新生血管、成纤维细胞的生长情况进行观察;愈合伤口数字化分析来进行评价。结果使用尼美舒利通过对COX-2信号通路进行调控对于皮肤伤口愈合有明显的影响,适当浓度的尼美舒利能够抑制伤口炎症反应,使成纤维细胞迁移控制在合适的程度。结论在小鼠手术伤口应用适当浓度的选择性COX-2抑制剂尼美舒利能减少病理性瘢痕的形成,值得进一步探讨其在创伤、美容、烧伤等方面的应用。 Objective To study COX-2 signaling pathway through vitro and vivo experiments,try to clarify the impact of COX-2 pathway to inflammatory cells in the skin wound,angiogenesis and evaluate the wound healing,in order to reduce scar formation,promote healing and provide experimental basis for diagnosis and treatment methods.Methods The scratch test in vitro cell culture was used,and in vivo experiments,COX-2 selective inhibitor Nimesulide intervening the COX-2 pathway was used and observed the inflammatory cells,angiogenesis,fibroblast cell migration through stretched skin and immunohistochemistry.The digital analysis of the scar was used to find a way to evaluate the healing process.Results Nimesulide had significant impact on COX-2 signaling pathways regulated the skin wound healing,the appropriate concentration of Nimesulide can inhibit wound inflammation and control the migration of fibroblasts at the right level.Conclusion Administration of Nimesulide(COX-2 selective inhibitor) with appropriate concentration has a positive effect on surgical wound in mice and could reduce scar formation.It is worthy of going further study on trauma,Cosmetic Surgery,Burn Surgery or other applications.
