摘要
目的探讨电针对食源性肥胖致不孕大鼠中枢性作用中,能否上调下丘脑弓状核GnRH的表达。方法将100只刚断乳雌性SD大鼠随机分为2组:①正常组20只,喂以普通饲料;②高能饲料组80只,给予高能饲料,筛选出肥胖不孕大鼠22只,并将其随机分成两组,即对照组和电针组。检测血清雌二醇(E2)、睾酮(T)、瘦素(leptin)、卵泡刺激素(FSH)、黄体生成素(LH)的水平;观察卵巢的形态学变化以及促性腺激素释放激素(GnRH)在下丘脑弓状核的表达情况。结果电针组较对照组大鼠血清E2、T、leptin水平明显降低,FSH、LH水平明显升高;下丘脑弓状核GnRH表达增强。结论针刺能够调节单纯食源性肥胖大鼠的神经内分泌代谢,从而降低由食源性肥胖所致的下丘脑-垂体-卵巢轴功能失调性无排卵而引起的不孕比例。
Objective To assess the central effect of electro-acupuncture (EA) on diet-induced obese infertility rats and explore if EA up-regulate the expression of GnRH in the arcuate nucleus(ARC) of the diet-induced obese infertility rats. Methods 100 female SD rats just weaned were divided into two groups. @The normal group with 20 rats were fed normal diet. @22 obese infertile rats were selected from the high-energy diet group with 80 rats and randomly divided into two groups, namely control group and electric acupuncture group. The levels of serum E2, T, leptin, FSH and LH in sera were evaluated. The morphological changes of ovarian and the expression of GnRH in arcuate nucleus of hypothalamus were observed. Results Compared with the control group, serum E2, T and leptin levels were decreased markedly in electric acupuncture group, whereas FSH and LH were significantly elevated. The expression of GnRH in arcuate nucleus of hypothalamus was increased (P 〈 0.05). Conclusion Acupuncture can modulate the neuroendocrine metabolism on simple diet-induced obese rats, thereby reducing the proportion of anovulatory infertility with the hypothalamus-pituitary-ovarian axis dysfunctional induced only by obesity.
