摘要
目的观测不同浓度芹菜素处理后基质金属蛋白酶-2及基质金属蛋白酶-9的分泌情况;检测CD44v6跨膜糖蛋白黏附分子的表达。方法酶谱分析法和酶联免疫吸附法测定不同浓度芹菜素处理后基质金属蛋白酶-2及基质金属蛋白酶-9的分泌情况;免疫组织化学法检测不同浓度芹菜素处理后细胞CD44v6的表达。结果酶谱分析法和酶联免疫吸附法测定均显示不同浓度芹菜素处理后基质金属蛋白酶-2及基质金属蛋白酶-9的分泌量随着药物浓度的升高而减少,呈时间和剂量。SW480细胞表达的CD44v6与芹菜素浓度呈负相关。结论芹菜素抑制SW480细胞分泌基质金属蛋白酶基质金属蛋白酶-2和基质金属蛋白酶-9,芹菜素是一种良好的基质金属蛋白酶抑制剂。芹菜素抑制SW480细胞粘附分子CD44v6的表达,这可能是它抑制肿瘤侵袭和转移的机制之一。
Objective To investigate the secretion of MMP-2 and MMP-9 and the expression of CIM4v6 in human SW480 cell after treated with apigenin. Methods Gelatin zymogram and enzyme-linked immunosorbent assay was used to analyze the effects of apigenin on production of matrix metalloproteinase-2 and 9. The protein expressions of CD44v6 in human SW480 cell after trea- ted by apigenin were measured by immunohistochemistry. Results Gelatin zymogram and enzyme-linked immunosorbent assay showed that secretion of matrix metalloproteinase-2 and 9 decreased after treated with apigenin. There was an inverse correlation between the protein expression of CD44v6 in human SW480 cells and dose of apigenin. Conclusion Apigenin could inhibit the secretion of MMP-2 and MMP-9 in SW480 cells, apigenin was a good matrix metalloproteinase inhibitors. Apigenin could suppress the protein expressions of CD44v6 on SW480 cells, It may be one of the mechanisms that it inhibited tumor invasion and metastasis.
