摘要
研究了影响CO2超临界流体萃取白芨组分的各种因素,获得了CO2流体萃取白芨组分的最佳工艺。以干粉减少量,萃取物质量和薄层层析(TLC)对萜类的检测作为萃取指标,以压力,温度,夹带剂种类和萃取时间作为影响因子,通过单因素和多因素正交设计实验,研究超临界萃取的优化实验条件。结果表明,单因素实验的适宜萃取温度为55℃,压力为25 MPa,夹带剂为无水乙醇,多因素正交实验表明,萃取温度为55℃,压力为20 MPa,萃取时间为1.5 h,夹带剂乙醇用量为干粉的1.5倍(g/mL比值)为最佳萃取工艺流程,在该条件下的萃取率为0.77%。
The factors which affected the supercritical CO2 fluid extraction for components from Bletilla striata were applied and the optimized process of the supercritical CO2 fluid extraction was obtained.The decrements of dry powder,the weight of extract,the detection of terpenoid compounds by thin-layer chromatography were used as extraction indicators.By taking the factors of the pressure,temperature,kinds of entrainer,times of extraction as impact factors,the studies on optimized conditions were carried out through single factor experiment and multivariate orthogonal design experiments in supercritical extraction.Results suggest that the suitable extraction temperature was 55℃,the pressure was 25 MPa and the entrainer was dehydrated alcohol in single factor experiments,the optimized extraction condition in multivariate orthogonal design experiments were 55℃ of temperature,20 MPa of pressure,1.5 hour of extraction time and 1.5 times of dosage of dehydrated alcohol(ratio of g/mL) and the extraction ratio was 0.77% in this condition.
