小鼠动脉粥样硬化斑块模型的快速制作及CD40在斑块中的表达 被引量:3

Establishing mouse model with atherosclerotic plaque and detecting expression of CD40 in the plaque lesions

摘要 目的探讨颈动脉硅胶圈植入法制作小鼠动脉粥样硬化(AS)斑块模型及斑块中白细胞分化抗原40(CD40)表达。方法 36只雄性载脂蛋白E(ApoE)-/-小鼠随机分为模型组和对照组:模型组小鼠采用颈动脉硅胶圈植入法快速制作AS斑块模型;对照组不予干预。苏木精-伊红(HE)染色检测斑块,免疫组化法检测斑块中CD40表达。结果模型组HE染色右颈动脉可见明显AS斑块形成,术后6周可见坏死中心区域和纤维帽形成,并且CD40表达阳性。结论颈动脉硅胶圈植入法能快速制作小鼠AS斑块模型,斑块中CD40高表达。 Objective To investigate the establishment of mouse model with atherosclerotic plaque and expression of CD40 in the plaque lesions.Methods Thirty-six male apolipoprotein E-deficient(ApoE-/-) mice were randomly divided into model group and control group.Silastic collars were placed around the right carotid arteries in model group,which were not used in control group.The slides of carotid arteries were stained routinely with hematoxylin and eosin(HE) and immunohis-tochemically with antibodies against CD40.Results Atherosclerosis formation on the right carotid arteries was seen in model group.The acellular necrotic core and well-defined fibrous cap were observed and the majority of these cells positively expressed CD40 in model group on the 6th week after surgery.Conclusion The collar-induced acceleration of carotid atherogenesis can be used to rapidly establish a mouse atherosclerosis model.CD40 is highly expressed in the atherosclerotic plaque lesions in the model mice with atherosclerosis.
