摘要
【目的】针对两个典型的水稻品种(系)耐盐材料(02-11和津源85)和两个敏感材料(秋田小町和99-28),进行水稻耐盐相关的SSR标记的研究。【方法】选取水稻12条染色体上的565对SSR引物进行亲本间多态性筛选;选择适合的标记引物,并确定PCR扩增的最适退火温度。【结果】在565对引物组合中筛选出68对引物在4个材料间有多态性;明确了主要标记引物的通用退火温度为55℃。【结论】68对可作为该水稻群体耐盐QTL分析的SSR标记引物,为进一步进行水稻耐盐性分子鉴定和耐盐分子辅助育种提供参考。
[ Objective ] The purpose of this study is to analyze the salinity - related SSR markers by applying SSR method to test the two typical salinity tolerance varieties ( 02 - 11 and Jinyuan 85) and the two typical salinity sensitive varieties (Qiutian Xiaoding and 99 - 28 ). [Method] A total of 565 polymorphic SSR markers on 12 chromosomes were applied to screen suitable primer on the four parents and the annealing temperature. [ Result] Sixty - eight pairs of SSR polymorphism primers were obtained from 565 pairs of SSR primers. It was confirmed that the suitable annealing temperature was at 55℃ . [ Conclusion] Sixty - eight pairs of SSR marker were used to analyze salinity- tolerance of rice varieties. It laid the foundation for molecular identify and molecular marker- assisted breeding of salinity - tolerance of flee.
