摘要
目的筛选出能够有效引起大肠杆菌ftsZ基因沉默的反义RNA片段。方法利用携带paired termini结构的质粒模仿天然asRNA的二级结构表达稳定的反义RNA,通过平板生长表象观察、菌体形态变化及液体培养生长速率变化考察了不同反义靶序列对大肠杆菌ftsZ基因沉默效果的影响。结果 ftsZ1、ftsZ4和ftsZ5 3个反义RNA片段可有效引起相关基因沉默。结论获得的ftsZ1、ftsZ4和ftsZ5 3个反义RNA片段可进一步用于ftsZ特异性酶抑制剂的超敏全细胞模型构建。
Objective To select the right antisense fragments with gene scilencing effects targeting ftsZ in Escherichia coli.Methods A vector with paired termini was to constantly express different asRNAs of ftsZ in Escherichia coli and the gene scilencing effects were observed by morphorlogy observation on plate,microscope and liquid culture.Results Three fragments ftsZ1,ftsZ4 and ftsZ5 were found inhibiting cell division effectively.Conclusions These cells with asRNAs can be further used as novel screenig models for selective ftsZ inhibitors.
