早期应用姜黄素对兔耳增生性瘢痕形成的影响 被引量:2

Effect of early curcumin treatment on formation of hypertrophic scar in rabbit ear

摘要 目的探讨早期应用姜黄素对兔耳增生性瘢痕形成的影响。方法选取10只健康的新西兰大耳白兔,体质量为2.5~3.0kg,手术制作兔耳创面,建立增生性瘢痕模型。将兔耳创面分为5组,每组24个创面。B组为空白对照组;术后第5天:注射生理盐水为TO组;分别于创面周围的皮下注射浓度为0.5mM(T1组)、1.0mM(他组)、2.0mM(乃组)的姜黄素,每个创面注射0.1ml,1次/d,注射至术后第4周。首先大体观察创面愈合时间和瘢痕增生情况,然后在瘢痕处取材,行HE、Masson染色,测量各组的HI指数,计算胶原纤维面密度,检测各组羟脯氨酸(HPr)含量及采用ELISA法测定血管内皮生长因子(VEGF)含量。结果①HI指数:T1组较B、TO组无明显变化,其差异无统计学意义(P〉0.05);T2、T3组较B、110组低,其差异有统计学意义(P〈0.05);T2、33组间比较,其差异无统计学意义(P〉0.05)。②胶原纤维面积密度:T1、,12、,13组低于B、rID组,其差异有统计学意义(P〈0.05);且以,12、,13组较低,但他、1r3组间比较,其差异无统计学意义(P〉0.05)。③Hpr含量:T1、仍组较B、11D组低,其差异有统计学意义(P〈0.05);且以T3组为最低。④蛋白水平:VEGF含量T1、他、仍组较B、11D组低,其差异有统计学意义(P〈0.05);且以他、13组较低,但,12、,13组间比较,其差异无统计学意义(P〉0.05)。结论在增生性瘢痕形成的早期阶段应用姜黄素能减少瘢痕组织中胶原的合成,减少瘢痕局部新生血管的生成,从而减轻瘢痕的增生,可为增生性瘢痕的临床治疗提供一定的理论依据。 Objective To investigate the effect of early curcumin treatment on formation of hypertrophic scar in rabbit ear. Methods Ten white health New Zealand rabbits with 2.5 ~3.0 kg were selected. The rabbit ear wounds were performed to establish the models of hypertrophic scar. The wounds were divided into 5 groups, each group with 24 wounds. Group B was as the control group ; Group TO was treated with NS at 5 clays postopera- tively ; Group T1, T2 and T3 were injected subcutaneously around wounds at 0.5 mM, 1 mM and 2 mM eurcumin concentrations resectively, 0.1 ml for each wound, once a day for 4 weeks. Healing time and hypertrophic scar were observed and then scar samples were harvested and performed HE and Masson staining. The HI indexes in each group, area density of collagen fiber, contents of hydroxyproline (Hyp) were detected and vascular endothe- lial growth factor (VEGF) was evaluated by ELISA. Results ① Compared with B and TO groups, HI index in T1 group had no change, there was no statistical significance ( P 〉 0.05) ; HI indexes in T2 and T3 groups were lower than those in B and TO groups and there was statistical significance ( P〈0.05), but there was no statistical significance between T2 and T3 groups ( P 〉 0.05 ). ②The area density of collagen fiber in the T1, T2 and T3 groups was lower than that in the B and TO groups, there was statistical significance ( P 〈 0.05) ; that was more lower in the T2 and 33 groups, but there was no statistical significance between T2 and 33 groups ( P 〉 0.05). ③ The content of Hyp in the T1 and 33 groups was lower than that in the B and TO groups, there was statistical significance ( P 〈 0.05) ; that was the lowest in the 33 group. ④ The content of VEGF in the T1, T2 and 33 groups was lower than that in the B and TO groups, there was statistical significance ( P 〈 0.05) ; that was more lower in the T2 and 33 groups, but there was no statistical significance between T2 and 33 groups ( P 〉 0.05). Conclusion Topical injection of curcumin at the early phase of hypertrophic scar formation can inhibit collagenous synthesis and reduce neovascular formation so as to lighten cicatricial hyperplasia. The findings of this study may have clinical implications on the management of human hypertryphic scars.
