缬沙坦对改善大鼠动脉血管内皮功能的研究

摘要 目的研究缬沙坦对离体大鼠主动脉内皮依赖性舒张的影响。方法选择老龄Wistar大鼠30只,随机分为老龄组15只,缬沙坦组15只,利用组织器官水浴系统,通过乙酰胆碱,测量离体主动脉环的舒张反应变化(%)。同时测定NOS在不同组大鼠主动脉中的含量。结果⑴老龄组主动脉环对乙酰胆碱(Ach,10-9~10-5 mol/L)诱发的血管内皮舒张反应均较缬沙坦组明显下降,两组之间差异具有显著性(P<0.01)。⑵在老龄组主动脉环血管内膜层可见棕褐色的反应颗粒,但与缬沙坦组相比较,可发现棕褐色的反应颗粒明显减少,NOS的分布下降降低(P<0.01)。结论随年龄增加,血管内皮细胞释放的NO逐渐减少,引起血管内皮舒张性降低,NOS随年龄的增加表达下降。与老龄组主动脉环相比,缬沙坦组主动脉环NO以及NOS的表达升高。
