摘要
日本三角涡虫是扁形动物门涡虫纲的代表动物,其独特的结构和极强的再生能力在研究胚胎发育和细胞分化中受到重视。但作为实验材料其个体较小,要取得足够量的组织存在困难,是研究工作中存在的问题。由于涡虫在受伤或被截断后,可以依赖体内的副胚层干细胞完整再生,因此,利用人工切割和饲养的方法可以培养大量的单克隆涡虫品系。研究目的在于建立涡虫单克隆群体的养殖体系标准,为后续的基因组和蛋白质组研究工作提供了依据。
It is essential to establish a standard culture system for monoclonal Dugesia japonica strains in the lab,since Dugesia japonica has been considered as a potential model in cell differentiation and embryogenesis studies due to its super regenerative ability.In this study,Dugesia japonica was cut into fragments,which then were cultured and regenerated to monoclonal groups.The monoclonal strain was consisted of 5 groups and the size of each group is from 37 to 45.The current technique for culturing Dugesia japonica may provide an useful tool for subsequent genomic and proteomic studies.
