高效毛细管电泳测定小果博落回中血根碱和白屈菜红碱 被引量:7

Content Determination of Sanguinarine and Chelerythrine In Macleaya microcarpa by HPCE

摘要 目的:建立测定小果博落回中血根碱和白屈菜红碱的含量的新方法。方法:采用高效毛细管电泳法,盐酸小檗碱为内标,运行缓冲液为25 mmol.L-1磷酸氢二钠与25 mmol.L-1磷酸二氢钠的混合体系(磷酸调pH为7.0)-甲醇(2.5∶1);运行电压18 kV;柱温25℃;检测波长200 nm。结果:血根碱和白屈菜红碱的线性范围分别为0.0124~0.0828mg.mL-1(r=0.999 8);白屈菜红碱的线性范围0.018 8~0.125 6 mg.mL-1(r=0.999 6),平均加样回收率(n=6)分别为98.7%(RSD1.01%)、98.4%(RSD 1.27%)。结论:高效毛细管电泳操作简便,分析快速,结果准确、重现性好,适用于小果博落回中血根碱和白屈菜红碱的含量的测定。 Objective:To establish a new method for determination of sanguinarine and chelerythrine in Macleaya microcarpa(Maxim).Method: Sanguinarine and chelerythrine in Macleaya microcarpa were determined by using high performance capillary electrophoresis(HPCE) with berberine as internal standard.The background electrolyte system selected was 25 mmol·L-1 disodium hydrogen phosphate and 25 mmol·L-1 sodium dihydrogen phosphate buffer at pH=7-methanol(2.5∶1);applied voltage,18 kV;temperature,25 ℃.Diode array detector was set at 200 nm.Result: The linear range of sanguinarine and chelerythrine was 0.012 4-0.082 8 g·L-1(r=0.999 8) and 0.018 8-0.125 6 g·L-1(r=0.999 6),The average recovery was 98.7%(RSD 1.01%) and 98.4%(RSD 1.27%).Conclusion: The method is simple,rapid and reproducible,and could be used for content determination of sanguinarine and chelerythrine In Macleaya microcarpa.
