摘要
目的:建立HPLC法同时测定草珊瑚中富马酸、异嗪皮啶和迷迭香酸含量。方法:采用Ultimate XB C18(250 mm×4.6 mm,5μm)色谱柱,流动相为乙腈(A)-0.1%磷酸溶液(B),梯度洗脱(0~10 min,2%A→20%A;10~35 min,保持20%A),流速1.0 mL.min-1,检测波长为208 nm,柱温为30℃。结果:富马酸,异嗪皮啶和迷迭香酸的线性范围分别为0.0190~0.133(r=0.9999),0.0204~0.143(r=0.9999),0.0212~0.148μg(r=0.9999);平均回收率(n=6)分别为101.3%(RSD=1.4%),99.1%(RSD=1.1%),98.2%(RSD=1.9%)。结论:该方法简便、快速、准确,适用于草珊瑚中富马酸、异嗪皮啶和迷迭香酸的含量测定。
Objective:To develop an HPLC method for simultaneous determination of fumaric acid,isofraxidin and rosmarinci acid in Sarcandra glabra.Methods:An Ultimate XB C18 column(250 mm×4.6 mm,5 μm) was adopted at 30℃.The mobile phases was acetonitrile(A) and 0.1% phosphoric acid water solution(B) with a gradient elution mode(0-10 min,2%A→20%A;10-35 min,20%A) at the flow rate of 1.0 mL·min-1.And the detection wavelength was 208 nm.Results:Fumaric acid,isofraxidin and rosmarinci acid were well separated with a linear range of 0.0190-0.133(r=0.9999),0.0204-0.143(r=0.9999),0.0212-0.148 μg(r=0.9999),respectively.The average recoveries were 101.3% with RSD 1.4%,99.1% with RSD 1.1%,98.2% with RSD 1.9%,respectively.Conclusion:The method is simple,rapid and reliable,and is suitable for content determination of fumaric acid,isofraxidin and rosmarinci acid in Sarcandra glabra.
