反相高效液相色谱法测定维药芜菁中槲皮素的含量

RP-HPLC determination of Quercetin in Brassica rapa L. for Uighur medicine

目的：建立HPLC法测定维药芜菁中槲皮素含量的方法。方法：反相高效液相色谱法,色谱条件：Kromasil C18柱（5μm,250 mm×4.6 mm）;流动相：甲醇-0.4%磷酸（42∶58,v/v）;检测波长：370 nm。结果：测定方法在5.0~50μg.mL-1浓度范围内呈良好的线性关系（r=0.9991）,平均回收率为99.2%（RSD=1.59%,n=6）。结论：方法简便可行、重复性好,可作为芜菁质量评价的方法之一。同时通过本实验测得芜菁中槲皮素的平均含量为203.8μg.g-1,此数据为进一步开发利用新疆丰富的药物资源提供了参考依据。

Objective：To describe a method for determining Quercetin in Brassica rapa L.by RP-HPLC.Methods：The determination was carried out over a Kromasil C18 column（5 μm,250 mm × 4.6 mm） at room temperature（20℃-25℃）.The mobile phase was CH3OH-0.4%H3PO4（42∶58,v/v） with flow rate of 1.0 mL· min-1,the detection wavelength was 370 nm.Results：There was a linear correlation between the concentration of Quercetin（5.0-50μg·mL-1） and the peak area value（r=0.9991）,the average recovery was 99.2% whith the RSD of 1.59 %（n=6）.Conclusion：The method is simple,and had good linear relationship.This method can be used to identify and evaluate the quality of Xinjiang Brassica rapa L..
