摘要
目的:建立HPLC法测定维药芜菁中槲皮素含量的方法。方法:反相高效液相色谱法,色谱条件:Kromasil C18柱(5μm,250 mm×4.6 mm);流动相:甲醇-0.4%磷酸(42∶58,v/v);检测波长:370 nm。结果:测定方法在5.0~50μg.mL-1浓度范围内呈良好的线性关系(r=0.9991),平均回收率为99.2%(RSD=1.59%,n=6)。结论:方法简便可行、重复性好,可作为芜菁质量评价的方法之一。同时通过本实验测得芜菁中槲皮素的平均含量为203.8μg.g-1,此数据为进一步开发利用新疆丰富的药物资源提供了参考依据。
Objective:To describe a method for determining Quercetin in Brassica rapa L.by RP-HPLC.Methods:The determination was carried out over a Kromasil C18 column(5 μm,250 mm × 4.6 mm) at room temperature(20℃-25℃).The mobile phase was CH3OH-0.4%H3PO4(42∶58,v/v) with flow rate of 1.0 mL· min-1,the detection wavelength was 370 nm.Results:There was a linear correlation between the concentration of Quercetin(5.0-50μg·mL-1) and the peak area value(r=0.9991),the average recovery was 99.2% whith the RSD of 1.59 %(n=6).Conclusion:The method is simple,and had good linear relationship.This method can be used to identify and evaluate the quality of Xinjiang Brassica rapa L..
