田口设计优化曲霉产多糖培养基

Optimization of culture medium for polysaccharide production by Aspergillus using Taguchi design

摘要 采用田口设计对多糖产量的培养基成分进行优化。先进行单因素试验,再利用Minitab软件设计正交试验,采用田口设计方法分析正交试验结果各因素水平的均值情况和信噪比情况。分析结果证明麸皮、硫酸镁和磷酸二氢钾对多糖产量影响极显著。软件预测培养基的最优组合为麸皮20g/L、酵母膏40g/L、MgSO4.7H2O0.25g/L和KH2PO41g/L,预测产量为8.86g/L,信噪比为20.1547dB。验证试验多糖产量为8.95g/L,信噪比为20.9213dB,优化方案达到了预期效果,具有较好的稳定性。 In this paper,culture medium for polysaccharide production by Aspergillus was optimized using Taguchi design.First,single factor was tested,then orthogonal experiment was designed to carry out using Minitab software,and the results of the mean and SNR of level of each factor were analyzed using Taguchi design.The results proved that the effects of wheat bran,magnesium sulfate and potassium dihydrogen phosphate on the polysaccharide yield were significant.Software predicts the optimal culture medium consisted of 20 g/L wheat bran,40 g/L yeast extract,0.25 g/L MgSO4·7H2O and 1 g/L KH2PO4,and the yield was 8.86 g/L,the SNR was 20.1547 dB.The polysaccharide yield of verification experiment was 8.95 g/L,and the SNR was 20.9213 dB.The results showed that the optimal solution achieved the desired effect with good stability.
