摘要
目的探讨抑癌基因早幼粒细胞性白血病基因(PML)在宫颈病变组织中是否存在突变。方法随机选取以下宫颈病变新鲜组织标本,包括宫颈上皮内瘤样病变I级(CINⅠ)、CINⅡ、CINⅢ、宫颈浸润鳞癌各30例、以及因其他疾病切除子宫病理证实为慢性宫颈炎者30例。用变性高效液相色谱(DHPLC)进行PML基因全部外显子及内含子-外显子交接区突变的筛查,对其中10例宫颈浸润鳞癌组织进行PML基因全部外显子及内含子-外显子交接区测序。结果全部组织均未发现PML基因突变。结论 PML在蛋白质水平的表达改变不是由其基因突变所引起。
Objective To study whether the tumor suppressor gene PML exists mutations in cervical lesions.Methods The following fresh tissue samples of cervical lesions were randomly selected,including 30 cases of CIN Ⅰ,CIN Ⅱ,CIN Ⅲ and cervical infiltrating squamous cell carcinoma respectively and 30 cases of chronic cervicitis.DHPLC was used to screen the PML exons and exon-intron boundaries for point mutations.We did PML exons and exon-intron boundaries direct sequencing for 10 cases of cervical infiltrating squamous cell carcinoma.Results We didn′t find any PML gene mutation in all samples.Conclusion PML expression change in protein level is not caused by the genetic mutation.
